麝亚百合与6个亚洲百合原种及栽培品种杂交亲和性及杂种后代增殖技术研究

【目的】探讨3个麝亚百合杂种系品种与6个亚洲百合原生种和栽培品种之间的杂交亲和性,以期培育出具有杂种优势的麝亚百合杂交后代。【方法】将3个麝亚百合品种(‘Benevento’‘Eyeliner’‘Couplet’)与6个源自亚洲的百合原生种及栽培品种(原生百合麝香百合Lilium longiflorum、岷江百合Lilium regale、鹿子百合Lilium speciosum var.gloriosoides Baker ‘Uchida’、大花卷丹Lilium leichtlinii var.maximowiczii,以及亚洲百合杂种系品种‘Calabria’‘Red Pearl’)进行远缘杂交,杂交后统计各杂交组合的果实膨大率和有胚率,对有胚种子进行胚珠离体培养以获得杂种苗。设置不同的萘乙酸(NAA)浓度以筛选出适合杂种后代不定芽增殖的最佳增殖培养基配方。【结果】3种麝亚百合做母本的杂交组合,只有‘Benevento’与6个百合父本都获得了膨大果实,且果实膨大率均≥80.00%;3种麝亚百合与大花卷丹的杂交亲和性最高,杂交后果实膨大率与平均有胚率均高于其他杂交组合,且均获得杂交后代;‘Benevento’×大花卷丹的杂种后代不定芽最适增殖培养基配方为MS+1 mg/L 6-BA+0.05 mg/L NAA+30 g/L蔗糖+7 g/L琼脂,pH 5.8~6.0。【结论】麝亚百合‘Benevento’适于作杂交的母本,而大花卷丹适于作杂交的父本;通过筛选最佳激素浓度配比,建立了杂种苗的最优增殖扩繁组培体系。该研究为今后麝亚百合远缘杂交育种提供了技术支持。

Abstract:

【Objective】This study aims to explore the hybrid affinity between three LA lilies and six native and cultivated varieties of Asian lilies, with the expectation of breeding LA lily hybrid offspring with hybrid vigor. 【Method】Three varieties of LA lily were subjected to distant hybridization with six original and cultivated varieties of lily from Asia. After hybridization, the fruit expansion rate and embryo formation rate of each hybrid combination were statistically analyzed. The embryo seeds were cultured in vitro with ovules to obtain hybrid seedlings, and then different NAA(naphthyl acetic acid) were set. The concentration of NAA was used to screen out the best proliferation medium formula suitable for the proliferation in hybrid offspring. 【Result】Among the hybrid combinations with three LA lilies as the female parent, only ‘Benevento' and six lilies as the male parent all obtained enlarged fruits, and the fruit enlargement rate was all greater than 80%. The hybridization compatibility between the three LA lilies and Lilium leichtlinii var. maximowiczii was the highest. After hybridization, the fruit expansion rate and the average embryo rate were both higher than those of other hybridization combinations, and all of them obtained hybrid offspring. The optimal proliferation medium formula for the adventitious buds of the hybrid offspring of ‘Benevento' × Lilium leichtlinii var. maximowiczii is MS+1 mg/L 6-BA +0.05 mg/L NAA +30 g/L sucrose +7 g/L agar, with a pH range of 5.8 to 6.0. 【Conclusion】It is found that LA lily ‘Benevento' is suitable as the female parent for hybridization, while Lilium leichtlinii var. maximowiczii is suitable as the male parent for hybridization. A tissue culture system for the proliferation and expansion of hybrid seedlings is established through the setting of hormone concentrations, which provides technical support for the distant hybrid breeding of LA lily in the future.
